Introduction to Colorectal Cancer and Gamma-Tocotrienol
CRC or colorectal cancer is one of the most dangerous types of cancer and is alone is the 3rd leading cause of cancer-related deaths in the US. The effects of Gamma-Tocotrienols on CRC cells were investigated both alone and in combination with capecitabine conditions in both vitro and in vivo studies. Cytotoxicity and Apoptosis assays were performed by FACS and MTT analysis. Western blotting and immunohistochemistry methods were used to investigate the expression of proteins. It was observed that Gamma-Tocotrienols inhibited the growth of human CRC and also sensitized CRC to capecitabine by regulating proteins linked to the growth of tumors.
How does Gamma-Tocotrienol fight Colorectal Cancer?
In the study, it was observed that Gamma-Tocotrienol inhibited the CRC proliferation with mutated KRAS. The Tocotrienol induced apoptosis, suppressed colony formation, and also suppressed key cell survival aiding regulators like an invasion, angiogenesis, cell proliferation, and metastasis.
It was also observed in the study that Gamma-Tocotrienol empowered the anticancer activities of capecitabine in CRC cells. Also, in a nude mouse xenograft model of human colorectal cancer cells, when Gamma-Tocotrienol was administered orally, the tumor growth was suppressed and the anti-tumor activity of capecitabine was enhanced. The immunohistochemical analysis and the western blot results showed that the expression of Ki-67, MMP-9, cyclin D1, CXCR4, VEGF, and Nf-kB was lower in tumor tissue from the combination treatment group.
